Mgr. Johannes Maria Trilaksyanta Pujasumarta lahir di Surakarta, 27 Desember 1949. Beliau adalah putra ketiga (dari sembilan bersaudara) dari pasangan Hubertus Soekarto Pudjasumarto dan Agnes Soekarti Pudjasumarto.
Ditahbiskan menjadi Imam pada tanggal 25 Juni 1977 dan terpilih menjadi Uskup Agung di Keuskupan Agung Semarang pada tanggal 12 November 2010. Salah seorang kakaknya juga menjadi imam, yaitu Romo Ignatius Ismartono SJ.
Selasa 10 Nopember 2015 pukul 23.30 WIB, telah tutup usia dengan tenang Uskup Agung Keuskupan Agung Semarang, mantan Uskup Diosis Bandung dan Sekretaris Jenderal KWI: Mgr. Johannes ‪Pujasumarta‬, setelah beberapa lamanya sakit, di RS Elisabeth, Semarang.
